    Antidepressants and Breast and Ovarian Cancer Risk: A Review of the Literature and Researchers' Financial Associations with Industry

    BACKGROUND: Antidepressant (AD) use has been purported to increase the risk of breast and ovarian cancer, although both epidemiological and pre-clinical studies have reported mixed results. Previous studies in a variety of biomedical fields have found that financial ties to drug companies are associated with favorable study conclusions. METHODS AND FINDINGS: We searched English-language articles in MEDLINE, PsychINFO, the Science Citations Index and the Cochrane Central Register of Controlled Clinical Trials (through November 2010). A total of 61 articles that assessed the relationship between breast and ovarian cancer and AD use and articles that examined the effect of ADs on cell growth were included. Multi-modal screening techniques were used to investigate researchers' financial ties with industry. A random effects meta-analysis was used to pool the findings from the epidemiological literature. Thirty-three percent (20/61) of the studies reported a positive association between ADs and cancer. Sixty-seven percent (41/61) of the studies reported no association or antiproliferative effect. The pooled odds ratio for the association between AD use and breast/ovarian cancer in the epidemiologic studies was 1.11 (95% CI, 1.03-1.20). Researchers with industry affiliations were significantly less likely than researchers without those ties to conclude that ADs increase the risk of breast or ovarian cancer. (0/15 [0%] vs 20/46 [43.5%] (Fisher's Exact test P = 0.0012). CONCLUSIONS: Both the pre-clinical and clinical data are mixed in terms of showing an association between AD use and breast and ovarian cancer. The possibility that ADs may exhibit a bi-phasic effect, whereby short-term use and/or low dose antidepressants may increase the risk of breast and ovarian cancer, warrants further investigation. Industry affiliations were significantly associated with negative conclusions regarding cancer risk. The findings have implications in light of the 2009 USPSTF guidelines for breast cancer screening and for the informed consent process

    Information retrieval and text mining technologies for chemistry

    Efficient access to chemical information contained in scientific literature, patents, technical reports, or the web is a pressing need shared by researchers and patent attorneys from different chemical disciplines. Retrieval of important chemical information in most cases starts with finding relevant documents for a particular chemical compound or family. Targeted retrieval of chemical documents is closely connected to the automatic recognition of chemical entities in the text, which commonly involves the extraction of the entire list of chemicals mentioned in a document, including any associated information. In this Review, we provide a comprehensive and in-depth description of fundamental concepts, technical implementations, and current technologies for meeting these information demands. A strong focus is placed on community challenges addressing systems performance, more particularly CHEMDNER and CHEMDNER patents tasks of BioCreative IV and V, respectively. Considering the growing interest in the construction of automatically annotated chemical knowledge bases that integrate chemical information and biological data, cheminformatics approaches for mapping the extracted chemical names into chemical structures and their subsequent annotation together with text mining applications for linking chemistry with biological information are also presented.     Development And Content Validation Of The Information Assessment Method For Patients And Consumers

    Background: Online consumer health information addresses health problems, self-care, disease prevention, and health care services and is intended for the general public. Using this information, people can improve their knowledge, participation in health decision-making, and health. However, there are no comprehensive instruments to evaluate the value of health information from a consumer perspective. Objective: We collaborated with information providers to develop and validate the Information Assessment Method for all (IAM4all) that can be used to collect feedback from information consumers (including patients), and to enable a two-way knowledge translation between information providers and consumers. Methods: Content validation steps were followed to develop the IAM4all questionnaire. The first version was based on a theoretical framework from information science, a critical literature review and prior work. Then, 16 laypersons were interviewed on their experience with online health information and specifically their impression of the IAM4all questionnaire. Based on the summaries and interpretations of interviews, questionnaire items were revised, added, and excluded, thus creating the second version of the questionnaire. Subsequently, a panel of 12 information specialists and 8 health researchers participated in an online survey to rate each questionnaire item for relevance, clarity, representativeness, and specificity. The result of this expert panel contributed to the third, current, version of the questionnaire. Results: The current version of the IAM4all questionnaire is structured by four levels of outcomes of information seeking/receiving: situational relevance, cognitive impact, information use, and health benefits. Following the interviews and the expert panel survey, 9 questionnaire items were confirmed as relevant, clear, representative, and specific. To improve readability and accessibility for users with a lower level of literacy, 19 items were reworded and all inconsistencies in using a passive or active voice have been solved. One item was removed due to redundancy. The current version of the IAM4all questionnaire contains 28 items. Conclusions: We developed and content validated the IAM4all in partnership with information providers, information specialists, researchers and representatives of information consumers. This questionnaire can be integrated within electronic knowledge resources to stimulate users' reflection (eg, their intention to use information).     A highly diverse siliceous sponge fauna (Porifera: Hexactinellida, Demospongiae) from the Eocene of north-eastern Italy: systematics and palaeoecology

    A siliceous sponge fauna, consisting of more than 900 specimens, is described from an early Lutetian tuffite horizon in the Chiampo Valley, Lessini Mountains, north-eastern Italy. Thirty-two taxa (15 Hexactinellida, 17 Demospongiae) are determined and illustrated, belonging to 24 genera, two of which are new (Rigonia gen. nov. and Coronispongia gen. nov.). Among these, 10 new species are proposed: Stauractinella eocenica sp. nov., Rigonia plicata gen. et sp. nov., Hexactinella clampensis sp. nov., Camerospongia visentinae sp. nov., C. tuberculata sp. nov., Toulminia italica sp. nov., Coronispongia confossa gen. et sp. nov., Cavispongia scarpai sp. nov., Corallistes multiosculata sp. nov. and Bolidium bertii sp. nov. Of the genera identified at Chiampo, 14 range back to the Cretaceous, three to the Jurassic and one to the Triassic, while six are still extant. The studied fauna shows affinities with sponges from the Eocene of Spain and the Cretaceous of Germany. The sponge fossils are uncompressed and bodily preserved, but the original siliceous skeleton is dissolved and substituted by calcite. Delicate attachments can be nevertheless documented: some sponges attached to a hard substrate by encrustation, while others were anchored on soft sediments by root-like structures. The presence of different modes of attachment suggests heterogeneous substrate conditions. Small, possibly young, sponges are recorded too. The sponge fauna is essentially autochthonous and lived in the middle-outer part of a carbonate ramp, where it formed clusters. This study extends the geographical and stratigraphical range of many sponge taxa, including Camerospongia, Toulminia, Ozotrachelus and Bolidium, previously documented only from the Cretaceous. The Recent calcified demosponge genus Astrosclera is reported here in the Cenozoic for the first time, having been reported previously in the Triassic only.
